High-density multicolour lineage tracing to address the clonal dynamics of epidermal cells over time is used. Rainbow3 transgenic mice expressing a Brainbow-1.0 construct under the control of the ubiquitous CAG promoter was crossed with Keratin 14-CreER mice. After tamoxifen-induced Cre recombinase activation, a transition from dTomato (expressed by default) towards random but stable expression of a combination of fluorescent protein genes occurred. Indeed, stable colour identities inherited by the cells’ progeny resulted over time in groups of cells that shared the same label. This allowed us to estimate the size and the location of clones originating from individual progenitor cells by cells harbouring the same colour. Photomicrographs represent 2D optical sections from a Z stack acquisition of a 0.5 mm skin section displaying IFE and HF harbouring multiple clones of different colours. (scale bar= 50 µm).
